Das Maus-FGF2-ELISA-Kit (Fibroblast Growth Factor 2, Basic) ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von FGF2.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus-FGF2 ist. Standards oder Proben werden den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, dann mit einem biotin-konjugierten Antikörper, der spezifisch für Maus-FGF2 ist. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen eine Farbänderung, die Maus-FGF2, einen biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus-FGF2 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
