Das Human-LPIN1 (Lipin 1) Mikrosample ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von LPIN1.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem für humanes LPIN1 spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, anschließend ein Biotin-konjugierter Antikörper, der spezifisch für humanes LPIN1 ist. Danach wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die humanes LPIN1, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, einen Farbwechsel. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und der Farbwechsel wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem LPIN1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
