Das Huhn-PGC (Pepsinogen C) ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Forschungsquantifizierung von PGC.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem Huhn-PGC-spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, danach ein Biotin-konjugierter Antikörper, der spezifisch für Huhn-PGC ist. Als Nächstes wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Huhn-PGC, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Huhn-PGC in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
