Der Human RASSF1 (Ras Association Domain Containing Protein 1) Mikrosample ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von RASSF1.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes RASSF1 ist. Standards oder Proben werden den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, dann ein Biotin-konjugierter Antikörper, der spezifisch für humanes RASSF1 ist. Als Nächstes wird Avidin, das mit Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen, die humanes RASSF1, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem RASSF1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
