Der Rind HSP-70/HSPA9 (Heat Shock Protein 70) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von HSP-70/HSPA9.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem spezifischen Antikörper gegen Rinder-HSP-70/HSPA9 vorbeschichtet. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für Rinder-HSP-70/HSPA9 ist. Anschließend wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die Rinder-HSP-70/HSPA9, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Rinder-HSP-70/HSPA9 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
