Das Humane GRAP2 (GRB2 Related Adaptor Protein 2) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsbestimmung von GRAP2.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem spezifischen Antikörper gegen humanes GRAP2 vorbeschichtet. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, gefolgt von einem Biotin-konjugierten Antikörper, der spezifisch für humanes GRAP2 ist. Als Nächstes wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die humanes GRAP2, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, einen Farbwechsel. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und der Farbwechsel wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem GRAP2 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
