Der Humane BMPR2(Bone Morphogenetic Protein Receptor 2) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von BMPR2.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes BMPR2 ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, danach ein biotin-konjugierter Antikörper, der spezifisch für humanes BMPR2 ist. Als Nächstes wird mit Meerrettichperoxidase (HRP) konjugiertes Avidin zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbveränderung, die humanes BMPR2, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und die Farbveränderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem BMPR2 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.

BMPR2 (bone morphogenetic protein receptor type 2; BMPR-II/BMPR-2) is a type II transmembrane serine/threonine kinase receptor in BMP signalling. In ligand-induced receptor complexes, BMPR2 activates type I BMP receptors, enabling downstream SMAD transcriptional responses.
Biological function: BMPR2 is a type II receptor serine/threonine kinase that binds BMP-family ligands including BMP2 and BMP7. Upon ligand-dependent receptor-complex formation, BMPR2 phosphorylates and activates type I BMP receptors; activated type I receptors then engage SMAD transcriptional regulators. Curated UniProt evidence also supports non-canonical signalling through the p38 MAPK cascade in defined contexts.
