Human ACTL6A (Actin Like Protein 6A) Microsample ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von ACTL6A.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes ACTL6A ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, danach ein Biotin-konjugierter Antikörper, der spezifisch für humanes ACTL6A ist. Anschließend wird Avidin, das mit Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die humanes ACTL6A, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem ACTL6A in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
