Der Human HCAM (Homing Associated Cell Adhesion Molecule) Mikrosample ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Erforschung und Messung von HCAM.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem spezifischen Antikörper für humanes HCAM vorbeschichtet. Standards oder Proben werden den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, gefolgt von einem biotin-konjugierten Antikörper, der spezifisch für humanes HCAM ist. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen eine Farbänderung, die humanes HCAM, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem HCAM in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
