Das Humane dLL1 (Delta Like Protein 1) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von dLL1.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem für Human dLL1 spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, dann ein biotin-konjugierter Antikörper, der spezifisch für Human dLL1 ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die Human dLL1, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, einen Farbwechsel. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und der Farbwechsel wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Human dLL1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
