Das Human PRM2(Protamin 2) ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Messung von PRM2 in der Forschung.
Das in diesem Kit angewandte Testprinzip ist ein Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem Human-PRM2-spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, anschließend ein Biotin-konjugierter Antikörper, der spezifisch für Human PRM2 ist. Danach wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Human PRM2, biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Schwefelsäurelösung gestoppt und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Human PRM2 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
