Der Human STX1A (Syntaxin 1A, Gehirn) Mikrosample ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Messung von STX1A in der Forschung.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für menschliches STX1A ist. Standards oder Proben werden den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, gefolgt von einem Biotin-konjugierten Antikörper, der spezifisch für menschliches STX1A ist. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die menschliches STX1A, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von menschlichem STX1A in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
