Der Human SEMA4D (Semaphorin 4D) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von SEMA4D.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem für humanes SEMA4D spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, danach ein biotin-konjugierter Antikörper, der spezifisch für humanes SEMA4D ist. Als Nächstes wird Avidin, das mit Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die humanes SEMA4D, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem SEMA4D in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
