Das Maus-CBG (Corticosteroid-bindendes Globulin) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von CBG.
Das in diesem Kit angewandte Testprinzip ist ein Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus-CBG ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, gefolgt von einem Biotin-konjugierten Antikörper, der spezifisch für Maus-CBG ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die Maus-CBG, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus-CBG in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
