Das Human OCLN (Occludin) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von OCLN.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem für humanes OCLN spezifischen Antikörper vorbeschichtet. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, anschließend mit einem Biotin-konjugierten Antikörper, der spezifisch für humanes OCLN ist. Als Nächstes wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die humanes OCLN, biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbveränderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbveränderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem OCLN in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
