Das Maus CXCR4 (Chemokin-C-X-C-Motiv-Rezeptor 4) Mikrosample ELISA Kit ist ein ELISA Kit von ELK Biotechnology zur Erforschung und Messung von CXCR4.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus-CXCR4 ist. Standards oder Proben werden dann zusammen mit einem Biotin-konjugierten Antikörper, der spezifisch für Maus-CXCR4 ist, in die entsprechenden Mikrotiterplattenvertiefungen gegeben. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die Maus-CXCR4, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus-CXCR4 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
