Der Simian C5b (Komplementfragment 5b) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von Simian C5b.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Simian C5b ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, anschließend ein Biotin-konjugierter Antikörper, der spezifisch für Simian C5b ist. Danach wird zu jeder Mikrotiterplattenvertiefung Avidin, konjugiert an Meerrettichperoxidase (HRP), gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen, die Simian C5b, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbveränderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbveränderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Simian C5b in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
