Tripeptidyl-Peptidase 1, auch bekannt als lysosomale Pepstatin-unempfindliche Protease, ist ein Enzym, das beim Menschen durch das TPP1-Gen kodiert wird. Dieses Gen kodiert ein Mitglied der Sedolisin-Familie der Serinproteasen. Die Protease funktioniert im Lysosom, um N-terminale Tripeptide von Substraten abzuspalten, und hat eine schwächere Endopeptidaseaktivität. Sie wird als katalytisch inaktives Enzym synthetisiert, das bei Ansäuerung aktiviert und autoproteolytisch gespalten wird. Mutationen in diesem Gen führen zu einer spätkindlichen neuronalen Ceroidlipofuszinose, die mit dem Versagen des Abbaus spezifischer Neuropeptide und einer Untereinheit der ATP-Synthase im Lysosom verbunden ist.
