TFPI (Tissue Factor Pathway Inhibitor), auch bekannt als EPI, LACI, TFI und TFPT1, ist ein einzelnes Polypeptid, das Faktor Xa (Xa) und Thrombin (Faktor IIa) reversibel hemmen kann. Während Xa gehemmt wird, kann der Xa-TFPI-Komplex anschließend auch den FVIIa-Tissue-Factor-Komplex hemmen. TFPI trägt signifikant zur Hemmung von Xa in vivo bei, obwohl es nur in Konzentrationen von 2,5 nM vorhanden ist. Das Gen für TFPI befindet sich auf Chromosom 2q31-q32.1 und hat neun Exons, die sich über 70 kb erstrecken. TFPI hat eine relative Molekülmasse von 34.000 bis 40.000, abhängig vom Grad der Proteolyse der C-terminalen Region.
