NOTCH4 (NOTCH, DROSOPHILA, HOMOLOG OF, 4), auch bekannt als INT3 oder NOTCH3 (ehemals), ist ein Mitglied der Notch-Familie. In-situ-Hybridisierung zeigte, dass Notch4-Transkripte primär auf Endothelzellen im embryonalen und erwachsenen Leben beschränkt sind, was auf eine spezifische Rolle von Notch4 während der Entwicklung des Wirbeltier-Endothels hindeutet. Die Sequenzen der Maus- und humanen NOTCH4-Proteine sind zu 82 % identisch. Northern-Blot-Analyse ergab, dass NOTCH4 (S) das Haupttranskript ist und in einer Vielzahl von Geweben exprimiert wird. Fluoreszenz-in-situ-Hybridisierung bestätigte die Lokalisation des NOTCH4-Gens auf Chromosom 6p21.3. Bei der Kopplungs-Disequilibrium-Kartierung der MHC-Region in 80 britischen Eltern-Kind-Trios fanden Wei und Hemmings (2000), dass NOTCH4 stark mit Schizophrenie assoziiert war. Die Repression von Notch4 löste Ataxie auf und kehrte den Krankheitsverlauf um, was zeigt, dass Notch4 nicht nur ausreicht, um die Krankheit auszulösen, sondern auch erforderlich ist, um sie aufrechtzuerhalten.
