Der Humane RARa (Retinsäure-Rezeptor Alpha) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsquantifizierung von RARa.
Das in diesem Kit angewendete Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem Human-RARa-spezifischen Antikörper vorbeschichtet. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, gefolgt von einem Biotin-konjugierten Human-RARa-spezifischen Antikörper. Anschließend wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Human-RARa, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten, einen Farbwechsel. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung gestoppt und der Farbwechsel spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Human-RARa in den Proben wird anschließend durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
