Das Human NGF (Nerve Growth Factor) Microsample ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Messung von NGF in der Forschung.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes NGF ist. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, danach ein Biotin-konjugierter Antikörper, der spezifisch für humanes NGF ist. Als Nächstes wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die humanes NGF, einen Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem NGF in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
