Das Huhn-KS(Keratansulfat)-ELISA-Kit ist ein ELK-Biotechnologie-ELISA-Kit zur Forschungsbestimmung von KS.
Dieser Assay verwendet die kompetitive Enzymimmunoassay-Technik. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit Hühner-KS vorbeschichtet. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, dann mit einem biotin-konjugierten Antikörper, der spezifisch für Hühner-KS ist. Anschließend wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Danach wird TMB-Substratlösung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Hühner-KS in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
