Das Human 8-OHdG(DNA-Schaden) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von 8-OHdG.
Dieser Assay verwendet die kompetitive Enzymimmunoassay-Technik. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit humanem 8-OHdG vorbeschichtet. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, danach eine biotin-konjugierte Antikörperlösung, die spezifisch für humanes 8-OHdG ist. Anschließend wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Danach wird TMB-Substratlösung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem 8-OHdG in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
