Das Rinder-MMP9 (Matrix Metalloproteinase 9) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsbestimmung von MMP9.
Das in diesem Kit angewendete Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Rinder-MMP9 ist. Standards oder Proben werden den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, gefolgt von einem Biotin-konjugierten Antikörper, der spezifisch für Rinder-MMP9 ist. Als Nächstes wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Rinder-MMP9, den Biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Rinder-MMP9 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
