Der Maus REG1a (Regenerating Islet Derived Protein 1 Alpha) ELISA Kit ist ein ELISA Kit von ELK Biotechnology zur Erforschung und Messung von REG1a.
Das in diesem Kit angewandte Testprinzip ist ein Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus REG1a ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, danach ein Biotin-konjugierter Antikörper, der spezifisch für Maus REG1a ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die Maus REG1a, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbveränderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbveränderung spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus REG1a in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
