Das Maus-TGFbI (Transformierender Wachstumsfaktor-Beta-induziertes Protein) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Untersuchung von TGFbI.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem Antikörper vorbeschichtet, der spezifisch für Maus-TGFbI ist. Standards oder Proben werden dann zusammen mit einem Biotin-konjugierten Antikörper, der spezifisch für Maus-TGFbI ist, in die entsprechenden Mikrotiterplattenvertiefungen gegeben. Als Nächstes wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Maus-TGFbI, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus-TGFbI in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
