Das Maus TYROBP (TYRO Protein Tyrosine Kinase Binding Protein) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsbestimmung von TYROBP.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Maus-TYROBP-spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für Maus-TYROBP ist. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die Maus-TYROBP, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus-TYROBP in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
