Technical note
Dieser ELISA-Kit nutzt das Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes IL-17C ist. Standards oder Proben werden zu den Vertiefungen der Mikro-ELISA-Platte gegeben und mit dem spezifischen Antikörper kombiniert. Anschließend werden nacheinander ein biotinylierter Detektionsantikörper, der spezifisch für humanes IL-17C ist, und ein Avidin-Meerrettichperoxidase (HRP)-Konjugat zu jeder Vertiefung der Mikroplatte gegeben und inkubiert. Ungestättigte Komponenten werden weggewaschen. Die Substratlösung wird zu jeder Vertiefung hinzugefügt. Nur jene Vertiefungen, die humanes IL-17C, den biotinylierten Detektionsantikörper und das Avidin-HRP-Konjugat enthalten, erscheinen blau. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet und die Farbe wechselt zu Gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von humanem IL-17C. Die Konzentration von humanem IL-17C in den Proben kann durch Vergleich der OD der Proben mit der Standardkurve berechnet werden.
