Dieser Assay-Kit basiert auf dem Sandwich-ELISA-Prinzip. Die PVDF-Membran am Boden der Mikro-ELISPOT-Platte wurde mit Anti-Maus-IFN-γ-Antikörpern neu beschichtet. Während des Experiments werden zuerst Zellsuspension und Stimulantien hinzugefügt. Das während der Inkubation von den Zellen sezernierte IFN-γ bindet an die beschichteten Antikörper. Anschließend werden die Zellen abgewaschen, und nacheinander werden biotinylierte Anti-Maus-IFN-γ-Antikörper und Alkalische Phosphatase-konjugiertes Avidin hinzugefügt. Die biotinylierten Anti-Maus-IFN-γ-Antikörper binden spezifisch an das bereits gebundene IFN-γ, und anschließend bindet Avidin spezifisch an Biotin, wodurch der Komplex gebildet wird. Nachdem freie Komponenten abgewaschen wurden, wird chromogenes Substrat (BCIP/NBT) hinzugefügt. BCIP wird unter Katalyse der Alkalischen Phosphatase hydrolysiert und reagiert mit NBT, wobei unlösliche, dunkelblaue bis blauviolette Spots gebildet werden. Eine professionelle ELISPOT-Plattenreader-Software kann zur Analyse der Ergebnisse verwendet werden, um die Frequenz der Maus-IFN-γ-sezernierenden Zellen zu berechnen.
