Dieser Assay-Kit basiert auf dem Sandwich-ELISA-Prinzip. Die PVDF-Membran am Boden der Mikro-ELISPOT-Platte wurde mit anti-humanen IL-5-Antikörpern rekotiert. Während des Experiments werden zunächst Zellsuspension und Stimulanzien hinzugefügt, und das von den Zellen während der Inkubation sezernierte IL-5 bindet an die beschichteten Antikörper. Anschließend werden die Zellen abgewaschen, und nacheinander werden biotinylierte anti-humane IL-5-Antikörper und alkalische Phosphatase-konjugiertes Avidin hinzugefügt. Die biotinylierten anti-humanen IL-5-Antikörper binden spezifisch an das bereits gebundene IL-5, und Avidin bindet dann spezifisch an Biotin, wodurch der Komplex gebildet wird. Nachdem freie Komponenten abgewaschen wurden, wird ein chromogenes Substrat (BCIP/NBT) hinzugefügt. BCIP unterliegt unter der Katalyse der alkalischen Phosphatase einer Hydrolyse und reagiert mit NBT, wodurch unlösliche Spots von dunkelblauer bis blauvioletter Farbe entstehen. Professionelle ELISPOT-Plattenlesesoftware kann zur Analyse des Ergebnisses verwendet werden, um die Frequenz der menschlichen IL-5 sezernierenden Zellen zu berechnen.
