Dieser Assay-Kit verwendet das Sandwich-ELISA-Prinzip. Die PVDF-Membran am Boden der Mikro-ELISPOT-Platte wurde mit anti-humanen IL-10-Antikörpern beschichtet. Während des Experiments werden zuerst Zellsuspension und Stimulanzien hinzugefügt. IL-10, das von den Zellen während der Inkubation sezerniert wird, bindet an die beschichteten Antikörper. Anschließend werden die Zellen abgewaschen, gefolgt von der sequenziellen Zugabe von biotinylierten anti-humanen IL-10-Antikörpern und Alkalische Phosphatase-konjugiertem Avidin. Die biotinylierten anti-humanen IL-10-Antikörper binden spezifisch an das bereits gebundene IL-10, und Avidin bindet spezifisch an Biotin, wodurch ein Komplex entsteht. Nachdem freie Komponenten abgewaschen wurden, wird ein chromogenes Substrat (BCIP/NBT) hinzugefügt. BCIP wird unter der Katalyse der Alkalischen Phosphatase hydrolysiert und reagiert mit NBT, wodurch unlösliche Flecken mit einer dunkelblauen bis blau-violetten Farbe entstehen. Professionelle ELISPOT-Plattenleser-Software kann zur Analyse des Ergebnisses verwendet werden, um die Frequenz der humanen IL-2-sezernierenden Zellen zu berechnen.
