Technical note
Dieses ELISA-Kit basiert auf dem Prinzip des kompetitiven ELISA. Die in diesem Kit enthaltene Mikro-ELISA-Platte wurde mit Kaninchen-Cortisol vorbeschichtet. Proben (oder Standards) und mit Meerrettichperoxidase (HRP) gekoppelte, für Kaninchen-Cortisol spezifische Antikörper werden in die Vertiefungen der Mikro-ELISA-Platte gegeben. Kaninchen-Cortisol in den Proben (oder Standards) konkurriert mit einer fixierten Menge an Cortisol auf dem Festphasenträger um die Bindungsstellen am HRP-gekoppelten, für Cortisol spezifischen Detektionsantikörper. Überschüssiges Konjugat und ungebundene Proben oder Standards werden von der Platte gewaschen. Die Substratlösung wird zu jeder Vertiefung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe der Stopplösung beendet und die Farbe wird gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 ± 2 nm gemessen. Die Konzentration von Kaninchen-Cortisol in den Proben wird anschließend durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
