Technical note
Dieses ELISA-Kit nutzt das Prinzip des kompetitiven ELISA. Die in diesem Kit enthaltene Mikro-ELISA-Platte wurde mit Affen-E2 vorbeschichtet. Proben (oder Standards) und Meerrettichperoxidase (HRP)-gekoppelte Antikörper, die spezifisch für Affen-E2 sind, werden in die Vertiefungen der Mikro-ELISA-Platte gegeben. Affen-E2 in Proben (oder Standards) konkurriert mit einer festen Menge an E2 auf dem Festphasenträger um Bindungsstellen am HRP-gekoppelten Detektionsantikörper, der spezifisch für E2 ist. Überschüssiges Konjugat und ungebundene Proben oder Standards werden von der Platte gewaschen. Die Substratlösung wird zu jeder Vertiefung gegeben. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet und die Farbe wird gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450±2 nm gemessen. Die Konzentration von Affen-E2 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
