Technical note
Dieses ELISA-Kit basiert auf dem Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit einem Antikörper vorbeschichtet, der spezifisch für Maus SELP ist. Proben (oder Standards) und mit Meerrettichperoxidase (HRP) gekoppelte Antikörper, die spezifisch für Maus SELP sind, werden in die Vertiefungen der Mikro-ELISA-Platte gegeben. Maus SELP in den Proben (oder Standards) bindet an den beschichteten Antikörper und an den HRP-gekoppelten Detektionsantikörper, der spezifisch für Maus SELP ist. Überschüssiges Konjugat und ungebundene Proben oder Standards werden von der Platte gewaschen. Die Substratlösung wird zu jeder Vertiefung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe der Stopplösung beendet, woraufhin die Farbe gelb wird. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von Maus SELP. Die Konzentration von Maus SELP in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
