Technical note
Dieser ELISA-Kit nutzt das Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit einem Antikörper vorbeschichtet, der spezifisch für Maus-ADP/Acrp30 ist. Proben (oder Standards) und Meerrettichperoxidase (HRP)-gekoppelte Antikörper, die spezifisch für Maus-ADP/Acrp30 sind, werden den Vertiefungen der Mikro-ELISA-Platte hinzugefügt. Maus-ADP/Acrp30 in Proben (oder Standards) verbindet sich mit dem beschichteten Antikörper und dem HRP-gekoppelten Nachweisantikörper, der spezifisch für Maus-ADP/Acrp30 ist. Überschüssiges Konjugat und ungebundene Proben oder Standards werden von der Platte gewaschen. Die Substratlösung wird zu jeder Vertiefung gegeben. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet, und die Farbe wird gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von Maus-ADP/Acrp30. Die Konzentration von Maus-ADP/Acrp30 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
