Technical note
Dieses ELISA-Kit basiert auf dem Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit einem Antikörper vorbeschichtet, der spezifisch für Maus-IgG2b ist. Proben (oder Standards) und mit Meerrettichperoxidase (HRP) gekoppelte, für Maus-IgG2b spezifische Antikörper werden in die Vertiefungen der Mikro-ELISA-Platte gegeben. Maus-IgG2b in den Proben (oder Standards) bindet an den beschichteten Antikörper und den HRP-gekoppelten Detektionsantikörper, der spezifisch für Maus-IgG2b ist. Überschüssiges Konjugat und ungebundene Proben oder Standards werden aus der Platte gewaschen. Die Substratlösung wird jeder Vertiefung zugegeben. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet, woraufhin sich die Farbe gelb färbt. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von Maus-IgG2b. Die Konzentration von Maus-IgG2b in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
