Technical note
Dieses ELISA-Kit basiert auf dem Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit einem Antikörper vorbeschichtet, der spezifisch für Maus-IgG1 ist. Proben (oder Standards) und mit Meerrettichperoxidase (HRP) gekoppelter Antikörper, der spezifisch für Maus-IgG1 ist, werden zu den Mikro-ELISA-Vertiefungen gegeben. Maus-IgG1 in den Proben (oder Standards) verbindet sich mit dem beschichteten Antikörper und dem HRP-gekoppelten Detektionsantikörper, der spezifisch für Maus-IgG1 ist. Überschüssiges Konjugat und ungebundene Probe oder Standard werden von der Platte gewaschen. Die Substratlösung wird zu jeder Vertiefung gegeben. Die Enzym-Substrat-Reaktion wird durch Zugabe der Stopplösung beendet und die Farbe wird gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450±2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von Maus-IgG1. Die Konzentration von Maus-IgG1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
