Technical note
Dieses ELISA-Kit verwendet das kompetitive ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte wurde mit Maus E3 vorbeschichtet. Proben (oder Standards) und Meerrettichperoxidase (HRP)-gekoppelte Antikörper, die spezifisch für Maus E3 sind, werden zu den Vertiefungen der Mikro-ELISA-Platte gegeben. Maus E3 in den Proben (oder Standards) konkurriert mit einer festen Menge E3 auf dem Festphasenträger um die Bindungsstellen am HRP-gekoppelten Nachweisantikörper, der spezifisch für E3 ist. Überschüssiges Konjugat und ungebundene Probe oder Standard werden von der Platte gewaschen. Die Substratlösung wird zu jeder Vertiefung gegeben. Die Enzym-Substrat-Reaktion wird durch Zugabe der Stopplösung beendet, und die Farbe wird gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 ± 2 nm gemessen. Die Konzentration von Maus E3 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
