Technical note
Dieses ELISA-Kit basiert auf dem Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit einem für humanes FABP3 spezifischen Antikörper vorbeschichtet. Proben (oder Standards) und mit Meerrettichperoxidase (HRP) gekoppelte Antikörper, die spezifisch für humanes FABP3 sind, werden in die Vertiefungen der Mikro-ELISA-Platte gegeben. Humanes FABP3 in den Proben (oder Standards) verbindet sich mit dem beschichteten Antikörper und dem HRP-gekoppelten Detektionsantikörper, der spezifisch für humanes FABP3 ist. Überschüssiges Konjugat und ungebundene Proben oder Standards werden von der Platte gewaschen. Die Substratlösung wird zu jeder Vertiefung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch die Zugabe einer Stopplösung beendet und die Farbe wird gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von humanem FABP3. Die Konzentration von humanem FABP3 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
