Technical note
Dieser ELISA-Kit nutzt das Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit einem Antikörper vorbeschichtet, der spezifisch für humanes TFF3 ist. Proben (oder Standards) und ein mit Meerrettichperoxidase (HRP) gekoppelter Antikörper, der spezifisch für humanes TFF3 ist, werden zu den Vertiefungen der Mikro-ELISA-Platte gegeben. Humanes TFF3 in den Proben (oder Standards) verbindet sich mit dem beschichteten Antikörper und dem HRP-gekoppelten Detektionsantikörper, der spezifisch für humanes TFF3 ist. Überschüssiges Konjugat und ungebundene Probe oder Standard werden von der Platte gewaschen. Die Substratlösung wird jeder Vertiefung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet, und die Farbe wird gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von humanem TFF3. Die Konzentration von humanem TFF3 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
