Technical note
Dieser ELISA-Kit nutzt das Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes HP ist. Proben (oder Standards) und mit Meerrettichperoxidase (HRP) gekoppelte Antikörper, die spezifisch für humanes HP sind, werden in die Vertiefungen der Mikro-ELISA-Platte gegeben. Humanes HP in Proben (oder Standards) verbindet sich mit dem beschichteten Antikörper und dem HRP-gekoppelten Detektionsantikörper, der spezifisch für humanes HP ist. Überschüssiges Konjugat und ungebundene Proben oder Standards werden von der Platte gewaschen. Die Substratlösung wird zu jeder Vertiefung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet, und die Farbe wird gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von humanem HP. Die Konzentration von humanem HP in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
