Dieser ELISA-Kit basiert auf dem Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit einem spezifischen Antikörper gegen Human C5a vorbeschichtet. Proben (oder Standards) und mit Meerrettichperoxidase (HRP) gekoppelter Antikörper, der spezifisch für Human C5a ist, werden in die Vertiefungen der Mikro-ELISA-Platte gegeben. Human C5a in Proben (oder Standards) verbindet sich mit dem beschichteten Antikörper und dem HRP-gekoppelten Detektionsantikörper, der spezifisch für Human C5a ist. Überschüssiges Konjugat und ungebundene Proben oder Standards werden von der Platte gewaschen. Die Substratlösung wird zu jeder Vertiefung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe der Stopplösung beendet, woraufhin sich die Farbe gelb färbt. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von Human C5a. Die Konzentration von Human C5a in den Proben wird anschließend durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
