Technical note
Dieses ELISA-Kit basiert auf dem Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit einem Antikörper vorbeschichtet, der spezifisch für humanes ADP/Acrp30 ist. Proben (oder Standards) und mit Meerrettichperoxidase (HRP) gekoppelte Antikörper, die spezifisch für humanes ADP/Acrp30 sind, werden in die Vertiefungen der Mikro-ELISA-Platte gegeben. Humanes ADP/Acrp30 in den Proben (oder Standards) bindet an den beschichteten Antikörper und den HRP-gekoppelten Detektionsantikörper, der spezifisch für humanes ADP/Acrp30 ist. Überschüssiges Konjugat und ungebundene Proben oder Standards werden von der Platte gewaschen. Die Substratlösung wird jeder Vertiefung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe der Stopplösung beendet, und die Farbe wird gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von humanem ADP/Acrp30. Die Konzentration von humanem ADP/Acrp30 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
