Technical note
Dieses ELISA-Kit basiert auf dem Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit einem für humanes IgG spezifischen Antikörper vorbeschichtet. Proben (oder Standards) und mit Meerrettichperoxidase (HRP) gekoppelter Antikörper, der spezifisch für humanes IgG ist, werden in die Vertiefungen der Mikro-ELISA-Platte gegeben. Humanes IgG in den Proben (oder Standards) verbindet sich mit dem beschichteten Antikörper und dem HRP-gekoppelten Detektionsantikörper, der spezifisch für humanes IgG ist. Überschüssiges Konjugat und ungebundene Proben oder Standards werden von der Platte gewaschen. Die Substratlösung wird zu jeder Vertiefung gegeben. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet und die Farbe wird gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration des humanen IgG. Die Konzentration des humanen IgG in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
