Technical note
Dieses ELISA-Kit verwendet das Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes Cys-C ist. Proben (oder Standards) und mit Meerrettichperoxidase (HRP) gekoppelte Antikörper, die spezifisch für humanes Cys-C sind, werden zu den Vertiefungen der Mikro-ELISA-Platte gegeben. Humanes Cys-C in Proben (oder Standards) verbindet sich mit dem beschichteten Antikörper und dem HRP-gekoppelten Detektionsantikörper, der spezifisch für humanes Cys-C ist. Überschüssiges Konjugat und ungebundene Probe oder ungebundener Standard werden von der Platte gewaschen. Die Substratlösung wird jeder Vertiefung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet und die Farbe wird gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von humanem Cys-C. Die Konzentration von humanem Cys-C in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
