Technical note
Dieses ELISA-Kit verwendet das Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes E-Cad ist. Proben (oder Standards) und mit Meerrettichperoxidase (HRP) gekoppelter Antikörper, der spezifisch für humanes E-Cad ist, werden in die Vertiefungen der Mikro-ELISA-Platte gegeben. Humanes E-Cad in Proben (oder Standards) verbindet sich mit dem beschichteten Antikörper und dem HRP-gekoppelten Detektionsantikörper, der spezifisch für humanes E-Cad ist. Überschüssiges Konjugat und ungebundene Proben oder Standards werden von der Platte gewaschen. Die Substratlösung wird zu jeder Vertiefung gegeben. Die Enzym-Substrat-Reaktion wird durch Zugabe der Stopplösung beendet und die Farbe wird gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450±2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von humanem E-Cad. Die Konzentration von humanem E-Cad in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
