Technical note
Dieser ELISA-Kit nutzt das Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit einem Antikörper vorbeschichtet, der spezifisch für humanes CEA ist. Proben (oder Standards) und mit Meerrettichperoxidase (HRP) gekoppelte Antikörper, die spezifisch für humanes CEA sind, werden in die Vertiefungen der Mikro-ELISA-Platte gegeben. Humanes CEA in Proben (oder Standards) verbindet sich mit dem beschichteten Antikörper und dem HRP-gekoppelten Detektionsantikörper, der spezifisch für humanes CEA ist. Überschüssiges Konjugat und ungebundene Probe oder Standard werden von der Platte gewaschen. Die Substratlösung wird zu jeder Vertiefung gegeben. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet, und die Farbe wird gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von humanem CEA. Die Konzentration von humanem CEA in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
