Technical note
Dieser ELISA-Kit basiert auf dem Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit einem Antikörper vorbeschichtet, der spezifisch für Human D2D ist. Proben (oder Standards) und mit Meerrettichperoxidase (HRP) gekoppelte Antikörper, die spezifisch für Human D2D sind, werden zu den Vertiefungen der Mikro-ELISA-Platte gegeben. Human D2D in den Proben (oder Standards) verbindet sich mit dem beschichteten Antikörper und dem HRP-gekoppelten Detektionsantikörper, der spezifisch für Human D2D ist. Überschüssiges Konjugat und ungebundene Proben oder Standards werden von der Platte gewaschen. Die Substratlösung wird zu jeder Vertiefung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe der Stopplösung beendet, woraufhin sich die Farbe gelb färbt. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von Human D2D. Die Konzentration von Human D2D in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
