Technical note
Dieses ELISA-Kit basiert auf dem Prinzip des kompetitiven ELISA. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit humanem E3 vorbeschichtet. Proben (oder Standards) und ein Meerrettichperoxidase (HRP)-gekoppelter Antikörper, der spezifisch für humanes E3 ist, werden in die Vertiefungen der Mikro-ELISA-Platte gegeben. Humanes E3 in den Proben (oder Standards) konkurriert mit einer festgelegten Menge an E3 auf dem festphasengebundenen Träger um die Bindungsstellen am HRP-gekoppelten Detektionsantikörper, der spezifisch für E3 ist. Überschüssiges Konjugat und ungebundene Probe oder Standard werden von der Platte gewaschen. Die Substratlösung wird jeder Vertiefung zugegeben. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet und die Farbe wird gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450±2 nm gemessen. Die Konzentration von humanem E3 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
